Thanks for your help. I found it out.
It gets broken together with the plugin wp-ulike (https://wordpress.org/plugins/wp-ulike/).
I didn’t find out why tough. Probably a jQuery issue.

Unfortunately I didn’t get any error. Everything looks good.
There’s also an EventListener which works. When I click I can see the highlights in the source code but the toggle of the class .closed is not working.
The only thing I could think of is the WP Version. We’re running WordPress 4.0.1. Maybe it could be a jQuery issue?

So, I checked debugging and it seems that there is nothing wrong with the flowplayer plugin.
The test website I’m using the plugin is .htaccess password restricted, but I think that shouldn’t be a problem? I’ve also tried it without password protection.
I think it’s somehow a javascript problem. I’ve tried latest browsers (Chrome, Firefox, Safari) and I get the same behavior on all browsers.
So now! When I manually remove the class .closed from the buttons (with developer tools in chrome) the box opens and the settings are there. Have you ever had a problem like that?
